Assessment of Toxicity of Intravitreal Triamcinolone Acetonide in Vitrectomized Rabbit Eyes

ABSTRACT
Purpose: We evaluated the toxicity of Triamcinolone acetonide (TAA) which was administered intravitreally to vitrectomized rabbit eyes in the different doses through different time periods by using electrophysiological and pathological tests.

Materials and Methods: Vitrectomy was performed in both eyes of 54 rabbits and different doses of TAA were injected intravitreally into the right eyes. Left eyes served as controls and received 0.2 ml. of balanced salt solution (BSS). Electroretinography was performed before surgery and at follow-up periods after TAA administration. Then the rabbits were sacrified at planned follow-up periods and the eyes were enucleated. Light microscopy examination was performed.

Results: During the 28-days period, 4 mg. TAA group never showed any toxicity sign in electrophysiological, histopathological and ophtalmological examinations. 6 mg and 8 mg TAA groups showed a decline at b-wave amplitude in ERG (especially in 8 mg group and at the late phases of 6mg group), and necrosis, atrophy, disorganisation, fibrosis at the retinal layers in varying degrees according to duration period and dose of the drug.

Conclusions: 4 mg triamcinolone acetonide can be administrated safely as an intravitreal application with its short effective half life and low side effect incidence. Doses above this limit may be very toxic to the retina. Keywords : Triamcinolone acetonide, vitrectomy, intravitreally, electroretinography
